Total Synthesis of Natural Products
Total Synthesis encompasses all aspects of organic chemistry: The development of new methodology, mechanistic and physical organic experiments, conformational and configurational analysis by spectroscopic and computational methods, and bioorganic and medicinal chemistry. Total Synthesis is also one of the broadest and most rewarding educational experiences for aspiring synthetic chemists.
Strain Release Chemistry
The release of molecular strain energy as a driving force in chemical transformations, often referred to as “strain-release” reactions, has led to unprecedented chemical transformations that provide access to novel scaffolds of interest to medicinal and material chemists. Our group has pioneered the chemistry of bicyclo[1.1.0]butanes (BCBs), one of the most strained (60-68 kcal/mol) carbocycles.
Heterocyclic Chemistry
More than half of all known organic compounds, and the vast majority of drugs, are heterocyclic. We are interested in the development of new stereoselective methods for the preparation of highly functionalized heterocycles. In particular, we have developed novel protocols for indole and five-membered heterocycle synthesis that have been widely adopted in industry and academe.
Medicinal Chemistry
We apply drug discovery and process chemistry methods for the structure-activity analysis of bioactive natural products and the preparation and scale-up of designed molecules. Specifically, we are involved in collaborative translational research with the goal of moving compounds into Phase I clinical trials. Some of these projects, such as flow and photo-flow synthesis and 3D printing, take advantage of the infrastructure of the Combinatorial Chemistry Center (CCC).
Computational Prediction of Macroscopic Properties
We are actively engaged in the computational assignment of the relative and absolute stereochemistry of organic molecules and the theoretical prediction of chiroptical phenomena and molecular and macroscopic properties, including structural features relevant for strain-release chemistry.
