Title: "Creating new inhibitors to SARS-CoV-2 Macrodomain using Fragments, Neutrons, and Entropy!"

Abstract: SARS-CoV-2  counteracts the immune response through the action of the nsp3 macrodomain, an enzyme that removes ADP-ribosylation marks from host proteins. This enzyme is a promising antiviral target because catalytic mutations render viruses nonpathogenic. I will discuss the discovery of  the first cell-active inhibitors for this protein and how several structural  technologies, including high throughput fragment screening, neutron crystallography, active learning, and ensemble modeling of protein conformational dynamics led to their discovery.
